Everything went right today! We were trying to remove a stabilizing tag from the protein, and it wouldn’t bind at all to our recovery column. Then, after we diluted it a bunch because we thought it was contaminated with salt (which it wasn’t), we needed to re-concentrate it. Then my partner dumped most of the 160 mL or so of protein solution onto the door of the freezer. What’s left (some was sucked off the freezer), we’re going to try again tomorrow with new columns, and hope all the mucking about hasn’t oxidized it all.
